چکیده

Periventricular nodular heterotopia (PNH) is a neuronal migration disorder characterized by nodules of gray matter located along the lateral ventricles, which can range from isolated single nodules to bilateral confluent nodules. This malformation of cortical development can be related to genetic or extrinsic factors. Reading dysfluency, epilepsy and normal intelligence are main hallmark. PNH and overlying cortex integrate a (dys)functional network. In this review, we discuss the recent progress about pathogenes and epileptogenesis on PNH, and their participation in the processing of higher cortical functions.

Periventricular heterotopia may result from radial glial fiber disruption.

Periventricular heterotopia (PVH) are collections of neurons and glia heterotopically located adjacent to the ventricles. The pathogenesis of periventricular heterotopia is believed to be a failure of cells to migrate from the ventricular zone. Mutations in filamin-1 (FLN1) have recently been identified as a genetic defect that results in an X-linked dominant form of PVH. In addition to this X-...

Abnormalities of cortical development and epilepsy

− The pre-natal administration of methylazoxymethanol acetate (MAM) in rats is able to induce cerebral heterotopia that share striking similarities with those observed in human periventricular nodular heterotopia, a cerebral dysgenesis frequently associated with drug-resistant focal seizures. In the present study, we investigated the mode of neurogenesis in cerebral heterotopia of MAM-treated r...

Infantile spasms with periventricular nodular heterotopia, unbalanced chromosomal translocation 3p26.2 ‐10p15.1 and 6q22.31 duplication

Patients presenting with infantile spasms, dysmorphic features, and periventricular nodular heterotopia may benefit from genetic copy number variation microarray, or whole-exome sequencing to identify candidate genes. This will allow personalized diagnosis and prognostication and the eventual understanding of single and combined gene functions in brain health and disease.

The role of periventricular nodular heterotopia in epileptogenesis.

A temporal resection in patients with periventricular nodular heterotopia (PNH) and intractable focal seizures yields poor results. To define the role of heterotopic grey matter tissue in epileptogenesis and to improve outcome, we performed stereoencephalography (SEEG) recordings in eight patients with uni- or bilateral PNH and intractable focal epilepsy. The SEEG studies aimed to evaluate the ...

Trouble making the first move: interpreting arrested neuronal migration in the cerebral cortex.

Postmitotic cortical neurons that fail to initiate migration can remain near their site of origin and form persistent periventricular nodular heterotopia (PH). In human telencephalon, this malformation is most commonly associated with Filamin-A (FLNa) mutations. The lack of genetic animal models that reliably produce PH has delayed our understanding of the underlying molecular mechanisms. This ...
